Project Abstract

Aim: The aim of this study was to investigate the expressions of Trophinin and CD26 (dipeptidyl peptidase IV) by immunoflourescence technique from cell adhesion molecules (CAM) in the existence of embryo from zygote to blastocyte stage in co-culture medium of received endometrial cells from mother applicants.

Materials and methods: Eleven women participated in the study whose ages varied from twentysix-to thirtysix years. Endometrial biopsies were received 21. day (luteal stage) of mentrual cyclus of women. Co-culture technique applied to endometrial cells. Controlled ovarian hyperstimulation (COH) applied to women. Oocytes were picked up with ultrasonography (US) after 2 days oocyte rupture (β hCG) injection was done. Intracytoplasmic sperm injection (ICSI) applied to selected oocytes in the embryology laboratory. Some of embryos obtained (n=25) were cultuvated in co-culture medium and some of them (n=80) were received in the convansional culture medium. Early developmental stages (1.-2.-3.-4.-5.days) of embryos were examined and good quality embryos were transferred in 5th day to the mother’s uterus by a catater. Endometrial cells were fixed in co-culture medium of transferred embryos. An immunohistochemistry procedure were applied to determine Trophinin and CD26 expressions in the endometrial cells. Trophinin and CD26 expressions were examined in endometrial co-culture cells by florescence microscopy.

Findings: At the end of this study, average of age was determined to be less significantly in pregnant group, compared to non-pregnant group (p=0.05). Number of Trophinin (+) stained cells was low significantly in pregnant group, in first day of embryos compared to non-pregnant group in endometrial co-culture (p=0.05). There was no significant difference in the number of CD26 (+) stained cells in existence of embryo in endometrial co-culture between the pregnant and non-pregnant groups.

Conclusion: Approximately half of the participant women in endometrial co-culture study were pregnant, the rest were not pregnant. We believe that immunohistochemical studies with co-culture medium will be useful to improve the convansional culture mediums. We consider that co-culture mediums have advantage over the communications with endometrial cells of embryos in early developmental stages.
